چکیده

Rat liver glycogen synthetase is strongly stimulated by cellular carboxylic acids which are intermediates of glycolysis and the tricarboxylic acid cycle. Activation of glycogen synthetase by the carboxylic acids may coordinate hepatic glycogen deposition with the functional status of the tricarboxylic acid cycle. At physiological concentrations of the carboxylic acids, the I form is preferentially stimulated. Whereas citrate, isocitrate, succinate, etc., stimulate total glycogen synthetase activity about 25 to SO%, these compounds stimulate the I activity 6to 8-fold. Citrate was used as a model compound in studies of the mechanism of glycogen synthetase activation. Stimulation of the enzyme is the result of modification of the glucose-6-P binding site which accompanies a conformational change mediated by citrate.

Regulation of rat liver glycogen synthetase. Relationship of the hormonal activation and the time-dependent in vitro activation.

The activation of glycogen synthetase by insulin proceeds via the transformation of the less active D form to the active I form, with essentially no change in total activity. Although the enhancement of glycogen deposition has also been attributed to glucocorticoid activation of glycogen synthetase, insulin-induced activation is counteracted by hydrocortisone in diabetic and starved rats. The t...

Regulation of hepatic glycogen synthesis during fetal development: roles of hydrocortisone, insulin, and insulin receptors.

In fetal rat liver in utero, an increase in glycogen and the glycogen synthetic enzyme glycogen synthetase (EC 2.4.1.11) occurs between gestational days 17 and 19. We used an organ culture system for finding the stimulus for increased enzyme activity and defining the relationship of this increase to glycogen synthesis. In fetal-liver explants from an earlier period (gestational day 16), hydroco...

Regulation of hepatic glycogen synthetase. Stimulation of glycogen synthetase in an in vitro liver system by insulin bound to sepharose.

Glycogen synthetase in an in vitro liver system was activated by Sepharose-bound insulin as well as by free insulin. The incorporation of uniformly CY4-labeled glucose into glycogen was also stimulated by either Sepharose-bound or free insulin. These results suggest that activation of glycogen synthetase by insulin is mediated by the interaction of insulin with the external cellular membrane an...

Regulation of hepatic glycogen synthetase of Rana catesbeiana.

A minced liver system from Rana catesbeiana tadpoles has been developed. Treatment of this liver preparation in vitro with insulin results in the activation of glycogen synthetase. Hormonally induced activation is the result of the conversion of the less active form of the enzyme to the more active form. The insulin-mediated activation is counteracted by hydrocortisone and dibutyryl-cyclic aden...

Hepatic glycogen synthetase deficiency

In the 13 years since hepatic glycogen synthetase deficiency was first described in identical twins no further cases seem to have been observed. We report a child who had suffered from occasional morning convulsions since the age of 7. Three 24-hour metabolic profiles showed fasting hypoglycaemia, hyperketonaemia, but normal lactate. Hyperglycaemia and hyperlactataemia occurred after meals. Glu...
